1) Constructing gene library of lipases and developing high throughput screening method for directed evolution of lipases;
2) Investigating the molecular mechanism of the thermo-stability of lipases;
3) Genetically engineering lipases by rational design and directed evolution towards higher catalytic activity, stability and substrate selectivity;
4) Establishing and fine-tuning the platform of the molecular evolution of enzymes;
5) Studying structure and function of lipases and exploring the molecular evolution mechanism of lipase family from sequence, structure to function;
6) Application assessment of modified lipases.

This project is to construct gene library of lipases and develop high throughput screening methods for lipase directed evolution, to explore the molecular evolution mechanism of lipase family from sequence, structure to function, to develop MutS based column or magnetic beads for the isolation and enrichment of mutants, to establish and fine-tune the platform of the molecular evolution of enzymes, with the final purpose of providing good enzymes for industries.
